dc.description.abstractBrassinosteroids (BRs)는 식물 호르몬 중 유일한 polyhydroxysteroids계 호르몬으로 세포 분열, 세포 성장, 관다발의 분화, 뿌리 생장 저해, 스트레스에 대한 내성 그리고 유전자 발현의 조절 등 다양한 생리적 작용에 관여한다 (Clouse and Sasse, 1998). BRs에 대한 연구가 활발히 이루어진 것이 얼마 되지 않아서 BRs의 세포 내 신호 전달 체계가 완전히 성립되지 않았다. 본 논문에서는 BRs의 복합 수용체의 일부인 BAK1에 결합함으로써 BRs의 세포 내 신호 전달 과정에 관여하는 물질을 찾기 위해 yeast two-hybrid screening을 수행하여 그 대상 후보 물질을 선택하고 그 물질에 대한 탐구를 수행하였다. BAK1에 결합하는 대상 후보 물질로 선택한 것은 glutathione S-transferase (GST) 중에서 GSTF10이라는 유전자로, E.coli를 이용한 pull down assay와 yeast를 이용한 coimmunoprecipitation를 통해 GSTF10이 만든 단백질이 실제로 BAK1과 결합을 하는지 알아보려 했으나 반복적인 실험에도 불구하고 결합을 확인할 수 없었다. 그래서 GSTF10이 BRs의 세포 내 신호 전달 과정에 특이적으로 관여하는 것으로 여겨지지 않는다. 그러나 GST가 BRs의 주된 역할인 식물의 성장과 관련된 생리적인 작용에 관여할 것이라 생각되므로 식물의 성장에 관여하는 것이라 알려진 여러 가지 호르몬과 스트레스를 처리하여 GST RNA 발현 정도를 알아보았다. 생장에 관여하는 호르몬인 GA_(3), IAA, BL를 처리하였을 때 GSTF10의 발현이 증가하는 것으로 나타났고, Me-JA, H2O2, SA, ACC를 처리하였을 시에도 발현이 증가하는 것으로 확인되었으나, ABA처리시에는 발현이 감소하였다. 또 저온과 가뭄 스트레스에서도 발현이 증가하는 것을 확인할 수 있었다. 따라서 BRs의 신호 전달 과정에서는 GSTF10의 관여를 단정지을 수 없지만 생리적인 과정에는 관여하고 있음을 확인할 수 있었으므로, BRs의 생리적인 과정 중 어떠한 역할에 GST가 관여하는지 알아보기 위해 GST 과발현 식물체와 RNAi 형질전환체를 제작하였다. 아직 T3 homozygote line을 선별하는 과정이라 정확한 표현형은 알 수 없다. 우선적으로 T2 세대에서 유전자 수준과 단백질 수준에서 GST 과발현으로 인한 그 기능을 유추하고자 GST의 과발현을 RT-PCR과 Northern blot analysis로 확인한 후 BRs에 따른 GST 활성을 확인하였다. 그 결과 GST 유전자가 증가하는 경향은 확인하였으나, 단백질 수준에서는 오히려 활성이 감소하였다. 그러나 과발현 식물체에서 GST 활성이 BRs 처리시 감소하는 정도가 야생형에 비해 줄어드는 것을 확인할 수 있었다. 이상의 연구들을 살펴보면, screening을 통해 BAK1에 결합하는 후보 물질로 선택한 GST 유전자가 BRs 세포 내 신호 전달 과정에 관여한다고 여겨지지는 않는다. 그러므로 GST외의 다른 clone들에 대한 연구가 수행되어야 한다. 반면, GST는 생리적인 작용에 전반적으로 관여하는 것으로 나타나므로 BRs의 생리적 작용 중 어떠한 부분에 관여하는지 정확한 기능을 알기 위해서는 현재 제작중인 형질전환체를 활용하여 추가적인 실험을 수행하여야 할 것이다.;Plant specific steroid hormone, brassinosteroids play essential roles in a variety of plant growth and developmental processes. Perception of brassinisteroids starts from the activation of receptor complexes consisting of BRI1 and BAK1, two different types of leucine rich repeats containing receptor-like serine/threonine kinases in Arabidopsis. After that little has been known about the very downstream signaling components involved in the BR signal transduction to regulate the BR-responsive gene expression, even though several proteins such as BIN2/DWF12/UCU1, BES1, BZR1 have been reported to mediate the BR signaling in the plant cells. This research was carried out to identify the downstream components of BRI1/BAK1 receptor complex. Yeast two-hybrid screening of expression library using the cytoplasmic kinase domain of BAK1 as a bait revealed the first 27 putative BAK1-interacting clones. 17 clones out of them turned out to encode a member of putative glutathione-S-transferases (GSTF10) which are ubiquitous in aerobic organism and catalyse the detoxification by conjugating the glutathione with toxic derivatives produced during the cellular processes. To find out whether GSTF10 and BAK1 actually binds, we performed pull-down assay and yeast coimmunoprecipitation. GSTF10 does not seem to be bound with BAK1. GSTF10 does not seem to be involved in the brassinosteroids signal transduction, but GST was involved in physiological actions in Arabidopsis. The expression of GST was induced by growth hormone of plants (GA, IAA, BL) and various abiotic stresses (cold, salt, drought). The transcription level of GST increased in GST overexpression plants, on the other hand the protein level decreased. But, the amount of decline of GST activity at BL treatment decreased in GST overexpression plants. The results in this research indicate that GST does not seem to be involved in the brassionsteroids signal transduction and appears to be involved in physiological actions, especially in growth and stress response.-
