dc.description.abstract신경능선유도체들은 분화의 방향이 결정되는동안에도 상당한 기간 가소성을 유지해서 다른 종류의 신경유도체로 교차분화(transdifferentiate)할 수 있다고 알려져 왔다. 특히 슈반세포는 멜라인세포와 근섬유모세포로 교차분화가능하다는 보고가 있었다. 그러나 세포분자수준에서 교차분화에 대한 심도깊은 연구는 충분한 양의 초기세포의 채취 어려움으로 인해 연구 진행이 어려운 상황이다. 본 연구에서는 슈반세포가 근섬유모세포로 교차분화하는 적절한 모델시스템을 제시하고자 한다. 특히, Sox10이라는 한가지 전사인자를 하향조절하는 전략을 이용해 이 현상을 유도했다는 점은 이 전사인자가 교차분화에 중요한 역할을 한다는 것을 암시한다.;Neural crest derived cells are noted for the long lasting plasticity during lineage commitment process and the potential for transdifferentiation into other neural crest derivatives. Schwann cells in particular have been reported to transdifferentiate into melanocytes and myofibroblasts. Detailed studies of transdifferentiation at the molecular level have been hampered by difficulty in isolating sufficient quantity of primary cells or cellular materials. Here, we describe a robust in vitro system in which Schwannoma cells undergo an apparent transdifferentiation into myofibroblasts. Importantly, we induce the transdifferentiation by downregulating a single transcription factor, Sox10, thereby identifying a key molecular event in this process. Myofibroblasts thus generated express several well-known markers of this lineage and contract in response to carbachol, a muscarnic against. These results suggest that generating desired cell types based on 'knock-down' of critical genes may be a viable strategy.-
