Abstract
MeCN-H₂O혼합용매에서 2,4-dinitrophenyl aceate와 p-chlorophenoxide, butane-2,3-dione monoximate와의 반응속도상수를 측정하였다. 기질에 대한 친핵체들의 반응성은 30mol % MeCN까지 MeCN을 첨가할수록 감소하고 그 이상으로 MeCN을 첨가하면 증가한다. 그리고 α-효과 (k^Ox^-/k^ClPhO^-)의 크기는 반응용매 중 MeCN의 mole %가 증가할수록 증가하였고, 이는 α-효과에 미치는 용매효과가 중요함을 의미한다. 다른 aryl acetates (p-nitrophenyl acetate, p-acetylphenyl acetate)와의 반응에서도 비슷한 결과를 얻었다. 용매 중 MeCN의 함량이 증가할수록 α-효과의 크기가 증가하는 이유는 MeCN함량이 증가할수록 각 친핵체들의 상대적인 염기도의 차이가 증가하였기 때문이었다. 물과 DMSO에서 일련의 이차 고리형 아민들과 p-nitrophenyl acetate와의 반응의 이차속도상수를 분광학적 방법으로 측정하였다. 물에서 DMSO로 용매가 변하였을 때 반응속도가 빨라졌다. 반응성에 미치는 용매 효과는 piperazinium ion과의 반응에서 가장 현저하였고 piperidine과의 반응에서 가장 작았다. piperazinium ion은 물에서보다 DMSO에서 1.04 pK_a만큼 염기성이 증가한 반면, piperidine은 DMSO에서 0.5 pK_a만큼 염기성이 감소하였다. 물과 DMSO에서 실행한 두 반응에서 Brønsted-type plot은 큰 기울기β값 (0.76~0.82)이 얻어졌으며, 이는 본 반응이 첨가 중간체가 분해되어 생성물이 생성되는 단계가 속도결정단계인 임을 의미한다. DMSO와 물에서 얻어진 pK_a값의 차이 (ΔpK_a)는 두 용매에서 얻어진 이차반응속도에서의 차이 (Δlog k_N)와 좋은 직선관계를 보이며, 이는 DMSO에서의 속도증가는 ΔpK_a의 차이에 의한 것임을 의미한다. 살충제의 일종인 paraoxone과 parathione의 염기촉매 가수분해 반응에 대한 이차속도상수는 반응용매 중 DMSO의 함량이 증가할수록 증가하였다. 그러나paraoxone의 반응에 대한 이차속도상수는 아래로 휘어지는 곡선을 보인 반면, parathione의 반응에 대한 이차속도상수는 위로 휘어지는 곡선을 나타내었다. 이런 상반되는 용매효과는 반응이 stepwise mechanism으로 진행한다는 증거로 볼 수 있었다. 치환된 phenylethynyl ketone과 일차아민과의 반응에서 모든 아민에 대하여 β_nuc값이 0.30±0.01인 직선의 Brønsted-type plot을 얻었으며, acid-base catalysis가 관찰되지 않았다. β_nuc값이 작다는 점으로부터 전이 상태에서 기질과 아민의 결합 형성이 적게 이루어짐을 알 수 있었다. 그리고 반응중심 탄소가 sp혼성화되어 있음에도 불구하고 α-효과의 크기는 10 정도로 작게 얻어졌는데, 이는 β_nuc값이 작았기 때문이다. 주어진 아민에 대하여 Hammett plot을 하였을 때 꺾인 직선의 그래프를 얻었다. 그러나 Brønsted-type plot은 모든 치환기에 대하여 비슷한 기울기를 갖는 직선의 그래프가 얻어졌다. Hammett 방정식 대신 Yukawa-Tsuno 방정식을 적용하여 보니 직선의 그래프를 얻을 수 있었다. Hammett plot에서 꺾임이 나타났던 것은 속도결정단계의 변화 때문이 아니라 전자주개 치환기를 가진 기질의 경우 resonance에 의해 바닥상태 에너지를 안정화시킬 수 있기 때문이다. 치환된 phenylethynyl ketone과 hydrazine을 반응시키면 enaminone을 거쳐 3-phenyl-pyrozole이 생성된다. enaminone이 생성되는 반응에서는 ρ값이 0.48인 직선의 기울기가 얻어져 치환기에 상관없이 같은 반응으로 진행함을 알 수 있었다. 그러나 enaminone에서 pyrazole을 생성하는 반응에서는 꺾인 Hammett plot이 얻어졌다. 이 Hammett plot을 기초로 전자밀개 치환기에서는 σ+값과 좋은 상관관계를 가지는 것으로 보아 질소원자가 carbonyl 탄소를 공격하는 단계가 속도결정 단계이고 전자끌개 치환기에서는 고리를 형성한 후 물이 빠져나가는 단계가 속도결정단계임을 알 수 있었다.;Second-order rate constants have been measured spectrophotometrically for the nucleophilic reactions of 2,4-dinitrophenyl acetate with butane- 2,3-dione monoximate and p-chlorophenoxide in MeCN-H₂O mixtures of varying compositions at 25.0±0.1℃. The reactivity of Ox^- towards the aryl acetates decreases upon addition of MeCN to the reaction medium up to ca. 30 mole % MeCN, followed by a gradual increase in rate upon further addition of MeCN. A similar result has been obtained for the reaction of ClPhO^- with the aryl acetates. Difference in the relative basicity of the nucleophiles is largely responsible for the increasing α-effect with medium composition in this system. .Second-order rate constants (k_N) have been measured spectro- photometrically for the reactions of p-nitrophenyl acetate with a series of alicyclic secondary amines in H₂O and in DMSO at 25.0±0.1.℃ Such a solvent change from H₂O to DMSO has resulted in rate enhancements. The effect of solvent on reactivity has been found to be most significant for the reaction with piperazinium ion while least significant for the reaction with piperidine. The Brønsted-type plot has been found to be linear with a large slope (nuc = 0.76-0.82) for both reactions run in H₂O and in DMSO, indicating that the aminolyses proceed through rate-determining break-down of the addition intermediate. The difference in pK_a values determined in DMSO and in H₂O has shown a linear correlation with the difference in the second-order rate constants determined in DMSO and in H₂O with a slope close to unity, suggesting that pK_a is fully responsible for the rate enhancement in DMSO. The second-order rate constant for the Alkaline Hydrolyses of paraoxone and parathione in DMSO-H₂O mixtures increases with increasing mole % DMSO in the reaction medium. However, the second-order rate constant for the reaction of paraoxone shows a downward curvature while that of parathione exhibits an upward curvature. The contrasting solvent effect profile obtained in the present study can be taken as evidence for a stepwise mechanism for the alkaline hydrolysis of paraoxone and parathione. The Brønsted-type plots are linear with small β_nuc values for the reactions of X-substituted phenyl ethynyl ketone with primary amines in H₂O. We expected a large α-effect for the reaction of hydrazine with the sp hybridized electrophile. However α-effect for the this reaction is small, and the small β_nuc values is considered to be responsible for the unusually small α-effect. The Hammett plots obtained are not linear but show a curvature as the benzyl substituent X becons electron donating ones for all amines studied, whereas the Brønsted-type plots are linear. The Yukawa-Tsuno plots obtained are linear, suggestinh that the nonlinear Hammett plots are not due to a change in the rate-determining step but due to resornance demand of the π-electron donor substituent on the benzyl moiety. Reactions with hydrazine have been found to give 3-aryl pyrazoles via enaminone. The UV spectral changes for the pathways suggested, i.e. absorbance at 340nm (enaminones) to reach a maximum increases initially and then decreases. A linear Hammett plot obtained with a slope of 0.48. and A nonlinear Hammett plot for the eneaminone formation reaction obtained, i.e., the slope decreases from 0.3 to near zero as the susbtituent changes from electron donating group to electron withdrawing ones. These results indicate that the RDS of the pyrazole formation process changes.
