dc.description.abstractBioinformatics는 생물학 연구 자료를 바탕으로 이로부터 새로운 생물학적 지식을 유추하기 위해 컴퓨터를 이용한 방법을 도입한 최신 과학 영역으로, genome sequencing project로부터 얻은 방대한 양의 생물학 자료를 분석하는 데에 결정적인 기여를 하고 있다. 청소년기 사람의 흉선 조직으로부터 T 세포군(TN, DP, SP)을 분리하고, PCR 방법을 통해 미량의 T 세포 RNA로부터 cDNA libray를 구축한 기존의 연구 결과가 있다. (고성호, 서울대학교 박사학위논문) 본 논문에서는 이러한 과정을 통해 얻어진 1386개의 EST들을 bioinformatics 및 computational biology 관점에 입각하여 기존의 프로그램 및 알고리즘을 활용한 재분석을 시도하였다. UniGene 및 SWISS-PROT 데이터베이스와의 local BLAST를 이용한 서열의상동성 비교 검색, 기능에 초점을 맞춘 motif 중심의 분석을 통해 각 EST 서열의 기능을 예측하였다. 즉, 기능이 연관된 여러 생물학적 서열들은 서로 비슷하며 같은 조상으로부터 유래되었을 가능성이 많다는 사실이 전제가 되는 상동성(homology) 검색 방법을 기반으로 하였다. 또한 T-cell의 발생 분화 단게에 따라 발현하는 유전자들의 발현 양상을 기능별로 7가지 카테고리로 나누어 분석하였다. 대량의 EST 서열을 조합하고 상동성 검색을 수행하는 과정에서 전산적 기법을 도입하였으며, 다양한 각도에서 유전자 서열 각각의 기능을 예측하고자 하였다. EST 서열은 그 자체가 매우 부정확함에도 불구하고, EST가 실존하는 genomic DNA의 일부분일 수 있기 때문에 이 서열 데이터의 분석은 의미가 있다. 본 연구에서는 지금까지 축적되어 온 생물학 정보 및 분석 프로그램들을 최대한 활용하였고 EST라는 데이터를 어떤 과정으로 분석하여야 할 것인가에 대한 전체적인 흐름을 제시하였다. 이는 생물학 연구의 새로운 방법이 될 수 있을 것이며, 논문에서 언급한 모든 절차가 하나의 web site 혹은 프로그램을 통해서 구현된다면 더 큰 효과를 기대할 수 있을 것이다.;Bioinformatics is the latest part of science using computer to analogize new biological knowledge from enormous biological research data. It contributes to analyze the experimental data that are the result of human genome sequencing project. The experiment was performed before this study : intrathymic T-cells for three developmental stages (TN, DP, SP) were isolated from the thymus of pre-adolescence child, and PCR-based cDNA libraries that are specific for each stage were constructed using cytoplasmic RNA extracted from the isolated thymocytes. (Sung-Ho Gho, Seoul National University) I attempt to reanalyze the 1386 ESTs obtaining from that procedure at a viewpoint of bioinformatics and computational biology using known programs or algorithms. I predicted the function of each EST sequence through sequence homology search with local BLAST of UniGene and SWISS-PROT database and functional motif-based analysis. The basis of those procedures are homology search method whose premise is that the biological sequences with related function are similar one another and are derived from the same ancestor. And I categorized the genes expressed following three T-cell developmental stages by 7 cellular functions. Here introduced the computational method in t he process of EST assembly and homology search, and attempted to predict the EST function from various viewpoints. Although themselves are very inaccurate, EST sequence analysis is important because ESTs can be the part of real genomic DNAs. In this study, I showed the possible flow for EST analysis, and tried to use utmostly both biological information and analytical tools accumulated till now. So, these procedure and result can be the new way of biological research. Furthermore, if those can be embodied in the web sites or aimple programs, biologists can expect better outcome of their research.-
