dc.description.abstractPeroxiredoxin (Prx) is a family of protein that reduces hydrogen peroxide (H2O2) using reducing equivalents of thioredoxin. The catalytic cysteine of Prx occasionally undergoes hyperoxidation to become cysteine sulfinic acid during catalysis, thereby leading to the inactivation of peroxidase activity. Prx III, a member of 2-Cys Prxs, is exclusively localized in the mitochondria. Mitochondria are the major source of H2O2, which can act as a second messenger and a crucial factor in cell functions involving cell signaling pathways. Mitochondrial Prx III is important to removing of mitochondrial H2O2, however, the mechanism of Prx III in cell signaling remain unclear. In this study, I demonstrate that mitochondrial Prx III redox regulation is related to cell signaling in peritoneal macrophages. In part I, I demonstrate that Prx III has a major role to play in the regulation of reactive oxygen species (ROS) and cell signaling in peritoneal macrophages. Macrophages produce ROS via NADPH oxidase 2 (Nox 2) during phagocytosis or stimulation with a wide variety of agents such as phorbol myristate acetate (PMA), PKC activator. The correlation between ROS derived from Nox 2 and Prx hyperoxidation in peritoneal macrophages and other cell types has not been reported. To find out whether ROS derived from Nox 2 has an effect on Prxs redox regulation, wild type and Nox 2 knockout (Nox 2-/-) peritoneal macrophages were treated with PMA. In the present study, I found that only Prx III was hyperoxidized in PMA-stimulated murine peritoneal macrophages independent of Nox 2. This hyperoxidation was blocked by the treatment of the cells with GF109203X, a PKC inhibitor. These results reveal that Prx III hyperoxidation is induced by PKC activation. To investigate whether H2O2 derived from Prx III hyperoxidation has a role in cell signaling, I checked the MAPK activation in PMA-stimulated peritoneal macrophages. ROS have been implicated in the activation of MAPK signaling pathways such as p38, JNK, and ERK. In peritoneal macrophage, p38 was activated by PMA. However, JNK and ERK were not activated. Also, I examined the mRNA level of cytokines, TNF-α, IL-1β, and IL-6, which are regulated by MAPK activation. The PMA-induced cytokine mRNA level had increased in the peritoneal macrophages. This data demonstrates that the accumulated H2O2 from Prx III inactivation activates p38 phosphorylation, which in turn induces cytokine release. In part II, I show that mitochondrial H2O2 derived from the Prx III inactivation has a central role in cell signaling. Mitochondrial Prx III acts independent of the cytoplasmic Prxs, however, its direct role in cell signaling such as MAPK activation remains unclear. To investigate the role of Prx III, I made Prx III Δ254 stable cell lines which express the c-terminal truncated Prx III mutant having immortal peroxidase activity. Prx III Δ254 cells had the lowest level of p38 activation by PMA stimulation. In contrast, the Prx III-depleted cell showed the highest level of p38 activation by PMA. I demonstrate that the H2O2 accumulation by Prx III hyperoxidation is important for MAPK signaling activation. In conclusion, these results of the present study suggest that the mitochondrial Prx III has a major role in redox signaling regulation of peritoneal macrophages. The PKC stimulated by PMA induces Prx III hyperoxidation, after which the accumulated H2O2 activates p38 MAPK and cytokine release. Based on these findings, I propose that Prx III is a critical target for cell signaling. ;페옥시레독신(Prx)은 과산화수소를 물로 전환하는 단백질의 구성원으로 이를 위해 사이오레독신을 환원당량으로서 사용한다. 촉매과정에서 Prx은 그것이 포함하는 촉매를 위한 시스테인의 잔기가 술핀산으로 전환되어 과잉산화 되면서 자신이 지니는 페록시다아제로서의 활성을 잃게 된다. 또한, Prx은 산화적 스트레스로 인한 신호들을 전환하는 산화환원 센서로서 작용한다. Prx 1과 2는 세포질에 존재하는 반면에 Prx 3은 예외적으로 미토콘드리아에 존재한다. 미토콘드리아는 H2O2를 생성하는 주요 세포소기관이며 이로부터 생성된 H2O2는 세포기능에서 세포신호전달 과정의 두번째신호전달자이자 중요인자로서 작용한다. 미토콘드리아에 존재하는 Prx 3은 미토콘드리아의 산화환원 세포신호전달과정에서 중요한 역할을 한다. 그러나 세포신호전달과정에서 Prx 3의 역할과 메커니즘은 명확하지 않다. 이번 이 연구에서는 복강대식세포에서의 Prx 3의 세포내부에서 산화환원과정과 관련된 세포신호전달과정을 살펴보고자 한다. 파트 1에서는 대식세포에서 활성산소종과 세포신호전달을 조절하기 위한 Prx 3의 주요역할을 살펴본다. 대식세포는 식균작용이나 PMA, 즉 PKC 활성체와 같은 다양한 시약에 의해 NADPH oxidase 2 (Nox 2)를 통해 활성산소종를 생성한다. 그러나 복강대식세포나 다른세포에서 Nox 2를 통해 유발된 활성산소종과 Prx의 관련성에 대해서는 알려진 바가 없다. Nox 2로부터 유발된 활성산소종이 Prx의 산화환원조절에 미치는 영향을 알아보기위해, 야생형과 Nox 2 가 제거된 (Nox 2-/-) 대식세포에 PMA를 처리하는 실험을 하였다. 그 결과 복강대식세포에서 PMA자극에 의해 생성된 활성산소가 Prx 3의 과잉산화를 유발했으며 이는 Nox 2와 관계없이 발생함을 관찰하였다. 이러한 과잉산화는 PKC 저해제인 GF109203X에 의해 저해됨을 관찰하였다. 이러한 결과는 Prx 3의 과잉산화가 PKC에 의해 조절됨을 보인다. 또한 이러한 Prx 3의 과잉산화로 인해 생성된 활성산소종이 세포신호전달에서 어떠한 역할을 하는지를 알아보기 위해 마우스의 복강세포에 PMA처리를 한뒤 MAPK의 활성화를 관찰하였다. 활성산소종은 p38, JNK, 그리고 ERK와 같은 MAPK의 세포신호전달과정에서 이들을 활성화시키는 것과 연관되어있음이 보고되어 왔다. 복강세포에서는 PMA에 의해 p38이 활성화 되었다. 그러나 JNK와 ERK는 PMA에 의해 활성화 되지 않았다. 또한, MAPK 활성화에 의해 조절되는 TNF-α, IL-1β, 그리고 IL-6와 같은 사이토카인의 mRNA의 양을 점검해 보았다. 복강세포에서는 PMA 자극에 의하여 사이토카인의 양이 증가되었다. 이 결과는 Prx 3의 불활성화에 의해 증가된 활성산소종이 p38의 인산화를 활성화하였고 이는 결국 사이토카인의 방출을 유발한 것임을 보여준다. 파트 2에서는 Prx 3의 불활성화로 인해 생성된 미토콘드리아로부터의 활성산소종이 세포신호전달과정에서 어떠한 역할을 하는지를 보인다. 미토콘드리아에 존재하는 Prx 3은 세포질에 존재하는 Prx들과는 독립적으로 세포내에서 활동하고 있으나 정확한 역할에 대해서는 아직까지 불분명하다. Prx 3의 역할을 알기위해, Prx 3의 돌연변이(Prx 3 Δ254), 즉 c-말단의 잔기가 제거되고 죽지 않는 페록시다아제의 활성화를 지니는 Prx 3을 발현하는 세포주 모델이 MAPK의 활성화에 영향을 미친다는 것을 보였다. Prx 3 Δ254를 발현하는 세포는 PMA자극에 대해 가장 낮은 p38의 활성화를 보였다. 이와는 대조적으로 Prx 3이 제거된 세포에서는 PMA에 의해 가장 높은 p38의 활성화를 보였다. 이는 Prx 3에 의해 조절되는 활성산소종이 MAPK의 활성화에 중요한 역할을 하고 있음을 보여준다. 결론적으로, 본 연구의 결과들은 미토콘드리아의 Prx 3이 복강대식세포에서 산화환원 신호전달과정의 주요한 역할을 한다는 것을 말해준다. PMA자극에 의해 PKC가 활성화되면 이는 Prx 3의 과잉산화를 유발하고 이로인해 축적된 활성산소가 p38MAPK와 사이토카인의 방출을 활성화한다. 이러한 발견을 기초로, Prx 3은 세포신호전달과정의 중요한 표적이 될 것으로 여겨진다.-
