- 해양 복족류인 총알고둥(Littorina brevicula)을 400μgIL 의 카드륨(CdCI₂)에서 30일 동안 노출시켜 cadmium-binding metallothionein (Cd-MT)을 유도하였다.
총알고둥의 소화선, 생식선, 아가미와 신장부위에서 세포 내 카드륨의 분포는 대부분(80%)이 용해된 상태로 세포질 단백질과 결합하고 있는 것으로 나타났다.
homogenized 조직의 supernatants 로부터 일련의 크로마토그래피 분리방법으로 Cd-MT가 정제되었다.
reverse-phase high-performance liquid chromatography (reverse-phase HPLC) 후, 하나의 Cd-MT peak 를 얻었다. 이 peak의 SDS-PAGE 분석 결과, 45kDa 근처에서 broad single band가 나타났다. 이 MT fraction 을 carboxymethylation 으로 sulfhydryl 기를 방해한 후에/는 apparent 분자량이 16kDa 인 하나의 band 를 관찰할 수 있었다. 많은 thiol기들 사이의 inter-disulfide 결합에 의해 단백질들이 뭉치게 된 것으로 사료된다. 이 MT는 mass spectrometric 분석을 통해 8.5kDa 의 분자량을 갖는다는 것으로 나타났다.;This research examines the induction of cadmium-binding metallothionein (Cd-MT) from the marine gastropod, Littorina brevicula, which was exposed to 400 ㎍/L Cd for 30 days.
The subcellular distribution of cadmium in combined tissues of the gill, digestive gland, gonad and kidney reveals that most cadmium(>80%) was bound to cytosolic (soluble) protein, metallothionein.
The Cd-MT was purified from supernatant of homogenized tissues, using various chromatographic techniques.
After reverse-phase high-performance liquid chromatography (reverse-phase HPLC), one Cd-MT peak was acquired. SDS-PAGE analysis of this peak showed a broad single band, around 45kDa. After blocking the sulfhydryl residues (carboxymethylation of MT fraction), we could observe only one band having apparent molecular weight 16kDa. We infer that inter-disulfide linkage between many thiol group led to MT aggregation. Mass spectrometric analysis revealed that the molecular masses of the MT were about 8.5kDa.
