Abstract
L-Glutamate는 중추신경계의 주요 흥분성 전달물질이다. 이것은 세포외액의 glutamate의 농도를 조절하는 excitatory amino acid transporters (EAATs)에 의해 일정한 농도로 유지된다. EAAT3 기능 장애는 신경 퇴행성 질환의 병리와도 관련이 있다. Clonidine은 선택적 α2 아드레날린 수용체 작용제로서 마취, 진정, 교감신경 차단, 진통 효과가 있어 마취과 영역에서 주목받고 있는 약물이다. Clonidine은 glutamate와 관련하여 신경보호 작용이 있음이 여러 논문을 통하여 알려진 바 있다. 이에 본 연구에서는 clonidine이 뇌에서 세포 외액의 glutamate를 조절하는 EAAT3의 활성도에 어떤 영향을 나타내는지 알아보고자 하였다. EAAT3를 발현시킨 Xenopus oocyte에 여러 농도 (7.50 × 10-10 - 1.88 × 10-7 M)의 clonidine을 전저치 한 후, L-glutamate (30 μM)를 적용시켜 two-electrode voltage clamp를 이용하여 막전위를 측정하였다. EAAT3의 Km, Vmax와 clonidine 효과의 가역성을 알아보기 위하여 다양한 농도의 L-glutamate (3-1000 M)의 존재 하에 1.50 × 10-7 M clonidine을 적용시키고 막전위를 측정하였다. Clonidine은 농도 의존적으로 glutamate에 대한 EAAT3 반응을 감소시켰으며, IC50 값은 2.72 × 10-8 M 이었다. 이 감소는 2.63 × 10-8 - 1.88 × 10-7 M 농도 범위에서 통계적 의미가 있었다. Clonidine은 1.50 × 10-7 M 농도에서 Vmax를 감소시켰으며, Km에는 영향이 없었다. 이 clonidine 효과는 Tyrode 용액을 4분 간 흘려준 후 약간 회복되었으나, 통계적 의의는 없었다. 본 연구는 clonidine이 임상에서 사용되는 농도에서는 EAAT3 활성도에 의미있는 영향이 없으나, 더 높은 농도에서는 EAAT3 활성도를 의미있게 감소시킨다는 것을 보여주었다. 이러한 결과는 clonidine의 진정 효과는 EAAT3와 관련이 없고, 오히려 임상농도 이상에서 보여진 EAAT3의 활성도 감소가 glutamate 과활성을 일으키므로 clonidine의 경련유발 기전을 설명할 수 있을 것으로 생각된다.;L-Glutamate is the major excitatory neurotransmitter in the central nervous system (CNS) and removed from the extracellular space by glutamate transporters (excitatory amino acid transporters, EAATs). Dysfunction of EAATs is specifically implicated in the pathology of neurodegenerative conditions. Clonidine, selective agonist of α2-adrenergic receptor agonist, exhibits anesthetic, sedative, sympatholytic and analgesic properties in clinical practice. It has been shown to be potent neuroprotectors related with the glutamatergic system. The aim of this study is to investigate the effects of clonidine on the activity of EAAT3 that can regulate extracellular glutamate. EAAT3 was expressed in the Xenopus oocytes. Using two-electrode voltage clamp, membrane currents were recorded after application of 30 μM L-glutamate in the presence or absence of various concentrations of clonidine (7.50 × 10-10 - 1.88 × 10-7 M). To determine the effects of clonidine on Km and Vmax of EAAT3 and reversibility of clonidine effects, membrane currents were recorded after application of various concentrations of L-glutamate (3-1000 μM) in the presence or absence of 1.50 × 10-7 M clonidine. Clonidine reduced EAAT3 responses to L-glutamate in a concentration- dependent manner with an IC50 value of 2.72 × 10-8 M. This inhibition was statistically significant at higher concentrations (2.63 × 10-8 - 1.88 × 10-7 M) than clinically relevant range. Clonidine at 1.50 × 10-7 M reduced the Vmax, but did not affect the Km of EAAT3 for L-glutamate. This clonidine effect disappeared slightly, but not significantly in oocytes perfused with Tyrode’s solution for 4 min after clonidine treatment. Our results showed that clonidine did not cause a significant change in the activity of EAAT3 at clinically relevant concentrations, but decreased EAAT3 activity significantly at higher concentrations. These results suggest that direct inhibition of EAAT3 activity is not related to sedation effect of clonidine and that clonidine-induced decrease of EAAT3 activity provides additional data for the possible involvement of a glutamatergic hyperactivity in the proconvulsant effect of clonidine.
