Abstract
식초는 동서양의 대표적인 발표식품으로 일상생활에서 조미료뿐 아니라 민간의약으로도 널리 사용되어 왔다. 특히 식초의 주요성분인 초산은 최근 연구에서 혈압 및 혈당조절, 지질대사 개선, 항 비만 등 다양한 기능성이 밝혀지고 있다. 이에 기능성을 강조한 식초음료 시장이 최근 급격히 성장 하고 있으나 아직까지 과학적 연구는 미흡한 실정이다. 따라서 본 연구에서는 현재 시판되고 있는 식초음료(홍초)의 항 비만 기능성을 검증하기 위해 홍초의 주요성분인 7 가지 종류의 과실식초에 대하여 HepG2 세포계를 통한 In vitro 유전자 스크리닝 및 in vivo 동물시험을 수행하였다. RT-PCR을 통한 7가지 과실 식초에 대한 in vitro 스크리닝 결과, 석류식초 처리 군에서 PPAR α, CPT-1, ACO 와 같은 지방산화 조절인자 및 효소의 유전자 발현이 유의적으로 가장 크게 증가되는 것을 확인 하였다. 더욱이 석류식초에 포함된 동일한 함량의 순수 초산과 비교 시, 석류식초 처리 군에서 동량의 초산 군보다 관련 유전자 수준이 유의적으로 증가하는 것을 확인하였다. 이는 식초의 주요성분인 초산뿐 아니라, 폴리페놀 등 과실식초가 가지는 추가 화합물들의 잠재적 기능성이 있을 것으로 기대된다. In vitro 연구 결과를 바탕으로 in vivo 모델에서의 생리학적 타당성을 검증하기 위해, 선정된 석류식초에 대하여 동물시험을 진행하였다. 수컷 Sprague-Dawley 쥐에게 41.2% 고지방식이로 비만을 유도하면서 석류식초 저 용량(VL), 고 용량(VH), 식초 섭취 군과 동일한 용량의 초산 저 용량(AL), 고 용량(AH)을 16주간 식이에 섞어 섭취하도록 하였다. 그 결과, 식초 또는 초산 섭취 군에서 대조군(C)에 비해 식이효율이 유의적으로 감소하였으며, VL 군과 AH군에서는 C군에 비해 체중증가가 유의적으로 억제되는 것을 확인하였다. 또한 VL군에서 부고환지방 및 신장주변지방이 유의적으로 감소되는 것을 확인하였다. VL군과 AH군에서 유의적 차이 없이 체지방 증가를 억제시키는 것으로 보아 석류식초에는 초산 외 체지방억제에 도움이 되는 다른 유용한 생리활성성분이 있을 것으로 기대된다. 더욱, 식초 군과 초산 군 모두 혈장 TG 및 LDL-콜레스테롤의 농도가 저하 된 것을 확인하였다. 이는 식초 및 초산이 지질대사개선에 긍정적인 효과를 보인다는 다른 연구결과와 일치하는 것이다. 또한 식초 및 초산 섭취군 모두 간에서 지방산화를 촉진하는 것으로 알려진 PPAR α와 CPT-1의 발현이 유의적으로 증가되는 것을 확인하였다. PPAR α는 간에서 인체 에너지 대사를 조절하는 AMPK 인산화에 의해 조절되는 것으로 알려져 있으며 선행연구에서 초산을 섭취시킨 쥐의 간 조직에서 AMPK가 인산화 되는 것이 확인되었다. 따라서, 식초는 간에서 지방산화 관련유전자들의 발현 증가를 통해 지방산화를 촉진함으로써 혈장 지질대사 및 항 비만에 긍정적인 효과가 있는 것으로 사료되며, 항 비만을 위한 기능성 식품소재로서의 가능성을 보여주었다. 하지만 지방, 근육 등 다른 조직에서의 추가적인 연구를 통해 관련 메커니즘이 연구되어야 하며, 더 나아가 사람을 대상으로 한 임상시험도 수행 되어야 할 것이다.;We investigate the anti-obesity effects of fruit vinegar which is major component of Hong Cho using in vitro and in vivo model. In vitro study, we screened the seven different kind of fruit vinegar obtained from DaeSang Corp. (Seoul, Korea) for the induction of fatty acid oxidation related genes in HepG2 cells using real-time PCR (RT-PCR). Among the fruit vinegars, red fruit vinegar mixed with pomegranate extract showed the highest mRNA expression level in peroxisome proliferator-activated receptor α (PPAR α), carnitine palmitoyl transferase-1 (CPT-1) and acyl-CoA oxidase (ACO) gene compared the control. According to this result, pomegranate vinegar seems to have positive effect on fatty acid oxidation and it may provide a possibility of anti-obesity effects. To test the physiological relevance of in vitro findings in vivo study was designed using the selected fruit vinegar. And possible dose-response relations of between the amount of acetic acid in the form of vinegar and pure acetic acid were compared in diet-induced obese rats. The animals were fed five different experimental diets supplemented with distilled water (Control; C), vinegar low-dose (VL), vinegar high dose (VH), acetic acid low-dose (AL) and acetic acid high-dose (AH) with 40% high-fat diet for 16 weeks. The body weight gain was significantly lower in VL, AH groups than the C group and food efficiency ratios were significantly lower in all treatment groups than the C group. The epididymal and perirenal fat pad was significantly lower in the VL group. Plasma TG and LDL-cholesterol content were significantly lower in all treatment groups than the C group. Hepatic mRNA levels of PPAR α and CPT-1 increase in the treatment groups compared to the C group. These result may have anti-obesity and plasma lipid lowering effects, which may be associated with upregulations of PPAR α and CPT-1 genes in the liver.
