dc.description.abstractIdentification of dendritic spine and their morphology change tracking in 2D time-lapse images play an important role in Neurobiology researches to examine the correlation between morphological characteristic and molecular regulation mechanism of spines. Moreover, one of the major issues is to find out the mechanism of genesis and development of dendritic spines. Recently, there are essential issues to resolve in related researches. Noises due to property of optical microscope disturb an accurate identification of dendritic spines and a temporal mapping to track the shape variation of spines efficiently. To solve these problems, in this paper we propose a spine detection technique robust to noise and micro-protrusion using structural information of neuron in 2D temporal optical microscopy images. Also we suggest an efficient local mapping method for tracking dynamics of dendritic spines in order to measure their morphological change quantitatively. First, we extract the tips in tree-like structure of dendritic spines as an initial contour position for deformable model based segmentation, and we use geodesic active contour model to detect the spines accurately. Second, we apply the optical flow method considering both the structure and the movement of objects to mapping every time-series image frames. We evaluate an accuracy of the proposed approach by comparing with the manual results, measurement results from SSEM and notable software NeuronIQ. In particular, this method is able to segment dendritic spines in noisy and non-uniform situations.;광학 현미경 기술의 발달은 살아있는 세포를 고배율로 관찰하는 것을 가능하게 하였다. 뇌 해마 신경세포의 관찰은 퇴행성 뇌질환의 진단을 가능하게 하며 신경세포의 단일 후시냅스 구획인 수상돌기 소극체(dendritic spine)의 구조는 기억, 학습, 그리고 알츠하이머 질환과 같은 퇴행성 뇌질환과 연관이 있음이 알려져 있다. 특히 수상돌기 소극체의 형태학적 변화는 시냅스의 신호전달 강도, 발생과 발달에 중요한 의미를 가진다. 따라서 형태학적인 구조에 따른 분자적 조절 메커니즘을 해명하기 위해 추출 및 시간 경과에 따른 변화를 추적하는 것이 필요하다. 그러나 광학 현미경은 명암도 분포가 고르지 않고 경계 부분의 번짐 현상 등으로 인해 정확한 경계 추출이 어려운 단점이 있다. 본 논문에서는 이러한 문제를 해결하기 위해 세포의 구조적 특성을 이용하여 잡음과 미세영역 추출에 강건한 분할 기법과 시간에 따른 수상돌기 소극체의 형태학적 변화의 정량적인 측정 기법을 제시한다. 기존의 방법들은 대부분 수상돌기 소극체의 전체 표면 정보를 이용한 분할을 수행하였으나 이는 명암도 문턱치화의 결과에 매우 민감하다. 따라서 본 논문에서는 수상돌기 소극체의 분기구조를 이용하여 변형모델기반 분할에 필요한 초기 윤곽선을 자동 추출하며 주변 잡음에 덜 민감한 변형모델 기반의 지오데식 동적 윤곽선 분할 기법을 적용하여 보다 정확한 소극체를 추출하고자 한다. 또한 시간에 따른 형태변화를 측정하기 위해 분할된 영역을 정량화하여 특징적인 3개의 종류로 분류한다. 시간에 따라 변화하는 수상돌기 소극체의 형태와 운동성을 추적하기 위해서는 영상 간 매핑이 필요하다. 기존 논문에서는 영상 전체에 대해 정합 기법을 이용하거나 골격을 이용한 그래프 매칭을 이용하여 영상간의 매핑을 수행하였다. 그러나 정합 방법은 영상의 작은 변형에도 뒤틀림이 발생할 수 있으며, 그래프 매칭 방법은 골격화의 결과에 의존적이다. 따라서 수상돌기 소극체의 가소성분석을 위해 시간에 따른 영상 간의 물체의 구조와 움직임을 고려하는 광학 흐름 기법을 이용하여 추적한다. 세포의 구조적 특징을 이용하여 관심영역 추출을 자동화하였으며, 변형모델에 기반한 수상돌기 소극체의 분할은 미세요철이 있는 돌출 추출에 효과적이다. 시간에 따라 빠르게 변화하는 수상돌기 소극체의 형태 및 운동성을 관찰하는데는 적합하지만 잡음과 불균일한 명암도분포를 갖는 이차원 영상에서도 관심영역을 효과적으로 분할한다. 또한 분할영역의 패턴분석 후 광학흐름 기반의 국소적 추적 기법 적용을 통해 시간 경과에 따른 형태 변화를 정량적으로 측정한다. 이 방법은 수동적인 측정 방법 및 기존의 자동화 방법, 그리고SSEM 영상에서의 측정 결과와 비교하여 그 성능을 평가하였다.-
