TIMP-2 -806T>C SNP have significant associations with the risk of endometriosis development in the Korean women

Abstract

Objective: To investigate whether single nucleotide polymorphisms and their haplotypes of gene encoding TIMP-2 are associated with the risk of endometriosis development in Korean women. Methods: In this case-control study, 254 endometriosis and 199 controls were enrolled. Blood samples were collected from Korean female patients with histopathologically confirmed endometriosis rAFS stage III and IV. The women undergoing laparascopic surgery or laparatomy for non-malignant lesions were included in the control group. The genetic distribution of TIMP-2 -418G>C polymorphisms were analyzed by polymerase chain reaction (PCR) and restriction fregment length polymorphism of PCR products. Other -806 T>C polymorphisms of TIMP-2 were analyzed by allele specific PCR. Their genotypes, allelotypes and haplotypes are analysed for evaluating the risk for endometriosis. Result: There was significant association between TIMP-2 806 T>C genotype and haplotype with the risk of endometriosis. However, TIMP-2 418 G>C polymorphisms showed no significant association with endometriosis. Conclusion: Our findings suggest that TIMP-2 -806 T>C polymorphisms are associated with risk of endometriosis development in Korean women.;본 연구에서는 TIMP-2의 -418G>C and -806T>C polymorphism이 자궁 내막증 발생 위험에 미치는 영향을 분석하였다. 254명의 자궁내막증 그룹과 199의 대조군이 등록되었고 자궁 내막증 그룹은 자궁 내막증 rAFS병기 III, IV기라고 조직학적으로 진단된 군이고 대조군은 내시경 수술 및 개복 수술에서 비악성 병변을 가진 그룹으로 제한하고 혈액 샘플을 채취하였다. TIMP-2 -418G>C polymorphisms의 유전학적 분포는 polymerase chain reaction (PCR)과 restriction fregment length polymorphism을 통해 분석되었고, TIMP-2 -806 T>C polymorphisms allele specific PCR을 통해 이루어졌다. 그들의 genotypes, allelotypes 그리고 haplotypes이 자궁 내막증 위험도를 평가하기 위해 분석되었다. TIMP-2 -418 G>C polymorphism은 wild type 동형접합자를 기준으로 하여 비교 위험도를 평가하였는데 variant genotype 동형 접합자와 이형접합자 모두의 위험도가 통계학적으로 의미 있지 않았다. 그에 반해 TIMP-2 -806 T>C polymorphism의 Genotype 빈도와 allelotype, haplotype은 모두 자궁 내막증 발현에 통계학적으로 의미 있게 높은 위험도를 보였다. TIMP-2 -418 G>C와 -806 T>C의 상관 관계를 보여주는Linkage disequilibrium 60% 이상으로 강한 상관관계를 가지고 있다고 볼 수 있다. 결론적으로 우리는 TIMP-2 -806 T>C polymorphism이 자궁내막증과 의미 있는 상관 관계가 있음으로 통계학적으로 증명 하였고 이 결과는 우리 나라 여성에서 자궁내막증 발현에 있어 TIMP-2 gene의 영향에 관한 증거가 되고 TIMP-2 -806 T>C의 새로운 기능에 관해 고찰할 수 있는 단서가 될 것이다.
