dc.description.abstractObjective: To investigate whether single nucleotide polymorphisms and their haplotypes of gene encoding CYP17 are associated with the risk of advanced endometriosis in Korean women. Methods: In this case-control study, 255 endometriosis and 199 controls were enrolled. Blood samples were collected from 255 Korean female patients with histopathologically confirmed endometriosis rAFS stage III and IV. The women undergoing laparoscopic surgery or laparotomy for non-malignant lesions were included in the control group. The genetic distribution of CYP17 27T>C polymorphisms were analyzed by polymerase chain reaction (PCR) and restriction fragment length polymorphism of PCR products. Other four common polymorphisms of CYP17 were selected to capture >95% of the variation in CYP17 gene and this included -12795G>A, -2618T>C, -1877C>T and -732A>G. Their haplotypes were also analysed for evaluating the risk for endometriosis. Result: There were no significant association between endometriosis and the genetic polymorphisms of -12795G>A, -2618T>C, -1877C>T, -732A>G and 27T>C. Logistic regression analysis indicated that the haplotype combinations of htSNPs were not significantly associated with the risk of endometriosis. Conclusion: Our findings suggest that CYP17 genetic polymorphisms including T27C are not associated with advanced stage endometriosis in Korean women.;목적 : 진행성 자궁내막증을 가진 한국여성에서의 CYP17 단염기 유전자 다형성과 haplotype간의 연관성을 살펴보기 위함. 방법: 이 환자 대조군 연구에서는 총 255명의 자궁내막증 환자군과 199명의 대조군이 포함되었음. 이들은 모두 조직병리학적으로 중증 자궁내막증(AFS stage III and IV)으로 확인되었음. 대조군은 에스트로겐 비연관성인 양성 부인과 질환으로 복강경이나 개복술을 시행받은 사람들을 포함하였음. CYP17 27T>C 다형성의 유전적 분포는 polymerase chain reaction (PCR)과 restriction fragment length polymorphism을 이용하였음. 나머지 CYP17의 대표적 단염기 유전자 다형성은 CYP17 의 다양성의 95% 이상을 보여줄수 있도록 선택되었음. 이들은 -12795G>A, -2618T>C, -1877C>T 과 -732A>G 로 이들의 haplotypes 또한 자궁내막증과의 연관성이 연구되었음. 결과 : 로그형 회기분석상 -12795G>A, -2618T>C, -1877C>T, -732A>G 과 27T>C로 대표되는 CYP17 단염기 유전자 다형성과 이들의 haplotype과 자궁내막증 사이에 유의한 연관성을 보이지 않았음. 결론: 본 연구결과 한국여성에서 T27C를 포함한 CYP17 단염기 유전자 다형성과 자궁내막증 사이에 유의한 관련성이 없음.-
