Abstract
BT(Bio Technology)의 한분야인 생명공학에서 줄기세포(stem cell) 연구 분야가 화제가 되었다. 세포의 정량적 측정은 이러한 세포연구에 있어서 그 연구 결과를 측정, 판단, 분석하는 중요한 지표가 된다. 이를 위하여 생물학자들은 경우에 따라 현미경 영상 속의 수많은 세포들을 직접 보고 세어야한다. 좀 더 현실적이고 자연적인 환경을 모방하여 실험을 하기위해 최근에 등장 하게 된 Hydrogel 과 같은 3-D micro environment 구조에서 세포수를 눈으로 세는 것은 더 힘이 든다. 이러한 과정에서 소비되는 많은 시간과 노력, 그리고 그 결과의 부정확성 확률에 대해서 생각해 보다면 이 과정의 자동화를 위한 연구는 중요한 의미를 가진다. 세포생존율이란 세포 성장의 정량적 측정의 한가지로 전체 cell sample 중, 살아있는 세포의 비율을 의미한다. 본 논문에서는 PEG-DA hydrogel 이라는 3-D 구조상의 hue7.5 세포에 대하여 컴퓨터 영상처리 기법을 사용하여 세포생존율을 구한 것을 생물학적인 방법으로 구한 세포생존율과 비교함으로서 성능을 평가하는데 그 목표를 두고 있다. 이를 위하여 생물학적인 세포생존율 테스트인 Live/Dead viability assay 와 alarmar blueTM Reduction를 수행하였고, 특히 Live/Dead viability assay에서 획득한 세포 영상에 대하여 세포생존율을 구하는 프로그램을 구현 하였다. 이 데이터 영상을 분석하여 영상 획득 실험 일, 샘플의 두께 등, 영상에 영향을 미칠 수 있는 요소에 따라 개선된 전처리를 하였고, 문턱치 정하는 방법 중에 Otsu's method 방법을 이용하여 영상을 이진화 하였다. 그리고 세포의 개수를 세기 위하여 각 이진화된 영역에 세포의 평균크기에 따른 가중치를 주어 그 값을 결정함으로서 세포생존율을 구할 시에 정확도를 높였다. 이렇게 구하여진 세포생존율은 생물학적인 방법으로 구한 세포 생존율과 비교해 볼 때, 평균 오차율 4.3%이내의 결과를 나타내었으며 PEG-DA hydrogel 3-d구조 기반의 hue 7.5 cell에 적용한 최초의 시도라는 의의가 있다. 또한 hue 7.5 세포를 연구하는 생물학자들이 세포생존율을 구하기 위해 소비하는 많은 시간과 노동력을 절약할 수 있게 하였다 그리고 더 나아가 직접 세포를 길러 획득한 영상 데이터를 가지고 영상처리를 통해 세포생존율을 구하고 이를 실제 직접 실험을 통해 얻어진 생물학적인 세포생존율과 비교했다는 점에서 보았을 때, IT와 BT 융합의 전문가적 지식을 가진 접근을 시도했다는 의미를 가진다. 앞으로 본 프로그램에 신뢰도를 높여 실제 상용화를 하는 것, hue7.5 세포 뿐 만이 아니라 다양한 세포의 세포생존율을 구할 수 있게 하는 것, 그리고 좀 더 세분화된 전처리와 영역추출을 통해 정확하게 세포의 분할이 가능하게 하는 것을 향후 연구 과제로 삼는다.;Tissue engineering research has the potential of bringing about significant discoveries in biology and medicine. Biologists often need to manually count the number of cells in conventional flat two dimensional (2-D) substrate (microwells) to study proliferating and divisional kinetics of cells. To better mimic the natural tissue structure, researchers often use Poly (ethylene glycol) di-acrylate (PEG-DA) hydrogels to generate a 3-D micro environment structure. Existing methods to study the viability of cell on this system are conventional 2-D cell viability tests such as live-dead assay, alamar blue. A major barrier of these methods to date is that the cell counting of the 2-D method does not represent the cell counting on the 3-D micro structure system. Biologists count cells manually, changing microscopy's focus hand-operatedly, while observing the 3-D micro structure system. That can have many limitations of accuracy and time consumption. The one way for improving this system is to place an automated cell counting method which could be a basic principle of cell viability analysis. We propose Cell viability analysis based on thresholding image segmentation considering cell size. Before thresholding segmentation, image preprocessing was done for good result. Among those threshold determination methods, we used Otus's method, a well-known effective segmentation method which is proposed by 'Otsu'. After that we calculate cell viability considering each cell size. The Basic conception steps of this thesis are the following; First, we will culture cells for getting data. Second, we will do two kinds of biological cell viability test which name are 'Live/Dead Viability Assay' and 'Alarmar BlueTM Reduction'. Third, for computer cell counting method, initial image data gets from "Live/Dead assay" which is two-color assay to determine viability of cells in a population using die. After counting number of cells using our method, we calculate viability by a mathematical formular. Then at the end, we will compare this result with biological cell viability test result.
