dc.description.abstractN-ethylmaleimide(NEM), which is experimentally used to alkylate the sulfhydryl group(s), has been shown to induce cytotoxicity in various kinds of cells such as hepatablastoma cells and lung cancer cell line (van Engeland et al., 1997;Kim et al., 2001). In primary astrocytes cultures, NEM concentration-dependently caused the release of LDH, a cell death or injury marker. The present study was aimed to investigate the mechanism for NEM-induced cytotoxicity in astrocytes. First, the oxidative stress-sensitive fluorescence dye,2,7-dihydro-dichlorofluorescein diacetate was used to study whether NEM provoked the formation of reactive oxygen species (ROS). The superoxide dismutase mimetics metalloporphyrins such as Mn(Ⅲ) tetrakis (N-methyl-4'-pyridyl) porphyrin (Mn-TMPyP) and Fe(Ⅲ)TMPyP did not inhibit the NEM-induced glial cell death, indicating that the ROS formation is not involved in the NEM cytotoxicity. Although NEM was previously reported to induce arachidonic acid release by activating cytosolic phospholipase A_(2)(cPLA_(2)), leading to ROS formation, the cytotoxicity of NEM was not inhibited by specific cPLA_(2) inhibitor, AACOCF_(3). Although it blocked the depolarization of mitochondrial transmembrane potential (MTP) in mitochondrial preparations, NEM induced the MTP depolarization in astrocytes. The MTP depolarization by NEM was completely blocked by the mitochondrial permeability transition (MPT) inhibitor cyclosporin A (CsA). However, CsA did not alter the NEM-induced gliad cell death, indicating the MTP depolarization by NEM might not play a crucial role in cell death. Treatment with NEM depolarized the plasma membrane potential (PMP). Removal of extracellular Na^(+), obtained by using Na^(+)-free medium, completely prevented both PMP depolarization and cell death caused by NEM. Removal of extracellular Cl^(-), obtained by using Cl^(-)-free medium, also decreased NEM-induced glial cell death but with no prevention of PMP depolarization. In contrast, the Ca^(2+) channel blocker CdCl_(2) prevented neither PMP depolarization nor NEM cytotoxicity. Further studies are needed to delineate He role of Na^(+) and Cl^(-) in NEM-evoked PMP depolarization and cell death.;N-ethylmaleimide(NEM)는 일반적으로 티올기를 알킬화하는 약물로서 세포내에 다양하게 분포하고 있는 티올기와 반응함으로써 다양한 약물작용을 일으키는 것으로 알려져 있다. 본 연구에서는 NEM이 신경 교세포(Astrocytes)의 사별을 유도한다는 것을 발견하였으며 NEM의 이러한 작용기전을 알아보고자 활성 산소종, 미토콘드리아, 그러고 이온농도의 변화와 관련하여 실험들을 진행하였다. 본 실험을 통하여 NEM 에 의해 세포내 활성 산소종의 증가는 보이네지 않았으며 아라티돈산 분비나 산화적 스트레스를 통해 NEM 이 교세포 사별을 유도하지는 않았다. 또한 NEM은 미토콘드리아 막전위의 탈분극을 유도하나 cyclosporin A라는 약물을 사용하여 탈분극을 막았음에도 불구하고 교세포 사멸이 진행되는 것으로 보아 NEM 에 의한 교세포 사멸 기전에 미토콘드리아 막 전위의 탈분극도 관련되어 있지 않음을 알 수 있었다. 마지막으로 본 연구에서 세포외부의 나트륨 이온과 염화 이온을 제지하여 NEM 에 의한 교세포 사별실험을 진행하였을 경우 NEM 에 의한 세포막 전위 탈분극과 세포 사멸이 나트륨 이온의 부재시 완전히 억제되었으며, 또한 염화 이온을 제거한 경우에도 세포 사멸이 크게 감소되는 것을 관찰할 수 있었다. 본 연구는 NEM 에 의해 신경 교세포 사별이 유도되며 이 기전에 나트륨과 염화이온이 관련되었을 가능성이 시사하고 있으며 NEM의 이온들의 역할은 앞으로 실험을 통해서 정확히 으로 보인다.-
