dc.description.abstract | To elucidate the pathogenic mechanisms involved in neurodegeneration in AIDS patients with cognitive deficits, the toxic effect of the lentivirus lytic peptide 1 (LLP-1) corresponding to the carboxyl terminus of HIV-1 transmembrane glycoprotein gp41 has examined on human neuronal and glial cell lines. LLP-1 induced a significant lactate dehydrogenase (LDH, a marker of cell death) release from these cells in a concentration and time dependent manner while the noncytolytic LLP-1 analog 2 had little effect. Application of LLP-1 to SH-SY5Y, a well-characterized human neuronal cell line, caused the decline of intracellular glutathione (GSH) content that appeared to occur before a significant LDH release. Furthermore, measurement of mitochondrial transmembrane potential (MTP) showed that LLP-1 elicited a significant loss of mitochondrial function. Among the reducing agents and antioxidants tested, GSH and a GSH prodrug N-acetyl cysteine (NAC) provided protection against LLP-1 induced neuronal cell death, evidently by restoring the intracellular GSH levels and blocking the disruption of mitochondrial integrity. Thus, gp41 derived LLP-1 may be a potential neurotoxic agent capable of causing the intracellular GSH depletion and disturbing the mitochondrial function, possibly contributing to the neurodegenerative cascade as seen in HIV-1 associated dementia. Our data indicate that restoring both GSH concentration and mitochondrial function may hold promise as possible therapeutic strategies for slowing disease progression of dementia in AIDS patients.;HIV-1 바이러스 구조 단백질의 하나인 HIV-1 바이러스 세포막 당 단백질인 gp41에 의한 뇌신경세포 사멸기전 양상을 고찰하고 이를 차단할 수 있는 억제제를 검색하고자 하였다. 그래서 HIV-1 gp41의 C-말단 lentivirue lytic peptide(LLP-1)에 의한 독성 실험을 인간 세포주인 SH-SY5Y와 교세포주인 T98G를 사용하여 LDH release assay와 위상차 현미경을 이용한 형태학적 검사를 수행하였다. 또한 신경세포독성과 연루된 기전을 분석하기 위하여 세포내 glutathione 농도 변화, 미토콘드리아 막전압 변화를 각 특이 형광색소를 이용하여 실험하였다. 첫째, LLP-1인 인간 신경 세포주인 SH-SY5Y에 미치는 세포독성이 시간/농도 의존성으로 유도되었고 교세포주인 T98G에서도 SH-SY5Y에 미치는 세포독성부다는 약하지만 거의 비슷한 패턴으로 시간/농도 의존성으로 유도되었다. 둘째, 신경 세포내에 존재하는 glutathione(GSH) 농도가 시간이 지남에 따라 LLP-1 처리군에서 점차 감소되는 것으로 확인되었고 미토콘드리아 막전압(MTP)도 시간이 지남에 따라 10□ LLP-1 처리군에서 점차 감소하는 것으로 확인되었다. 셋째, LLP-1에 의해 초래되는 신경 세포 독성이 GSH와 GSH prodrug NAC에 의해 유의하게 억제되었고 미토콘드리아 기능 변화 즉 MTP의 감소도 유의하게 억제되는 것으로 확인되었다. 따라서 본 연구논문은 HIV-1 바이러스 감염환자에서 발생하는 치매를 포함하는 토생성 뇌질환에 있어 HIV-1 바이러스 단백질중 gp41-LLP-1이라는 단백질이 신경세포 및 성상세포의 세포사를 유발하고 면역세포들의 활성화와 과다 염증 반응시 나타날 수 있는 간접적인 신경세포 변성 과정과의 유의성을 제시하는 자료로도 충분히 의미를 둘 수 있다. 또한 GSH와 NAC 등과 같은 LLP-1 독성 억제제가 LLP-1에 의한 신경세포내 GSH농도 감소 및 미토콘드리아의 기능 변화 즉 MTP감소를 차단 할 수 있다는 새로운 사실을 밝히고 있다. 이러한 것들은 AIDS환자들에서 치매 질환의 진행을 경감하거나 차단할 수 있는 치료적인 효과들을 가능하게 할 것이다. | - |
