Abstract
중간엽 줄기세포는 골수 기질 세포의 하나로 조혈작용을 돕는 것과 더불어 다양한 중간엽 세포로 분화할 능력을 가지고 있다. 또한, 최근 중간엽 줄기세포가 단구 기원의 수지상 세포의 분화를 억제함으로써 면역억제 능력이 있음이 밝혀졌다. 수지상 세포는 림프절로 이동하여 T세포에 항원을 제시하는데, CCL21은 림프절의 T세포 구역으로 수지상 세포를 이동시키는 중요한 chemokine이다. 본 연구에서 마우스의 수지상 세포를 이용하여 중간엽 줄기세포가 수지상 세포의 성숙에 있어 특징적인 화학 주성 작용에 미치는 영향을 알아보았다. 골수 세포를 분리하여 GM-CSF, lipopolysaccharide를 첨가하여 골수계 수지상 세포를 배양하고 이때 일부는 중간엽 줄기세포와 함께 배양하고, 대조군은 중간엽 줄기세포 없이 배양하였다. 중간엽 줄기세포를 면역 표지자를 이용한 유세포 분석과 colony forming unit-fibroblast assay를 통해 확인하였다. RT-PCR로 수지상 세포의 chemokine receptor 발현을 확인하였다. 수지상 세포의 이동은 새롭게 개발된 Time-lapse video microscopic technigue로 관찰하였다. 중간엽 줄기세포와 수지상 세포를 함께 배양한 결과 수지상 세포의 초기 분화 뿐 아니라 성숙도 억제되었다. 성숙된 수지상 세포는 수지상 세포의 강력한 chemokine인 CCL21에 반응해 활동적으로 이동한 반면 중간엽 줄기세포와 함께 배양한 세포는 그렇지 못했다. 이 실험은 중간엽 줄기세포가 수지상세포의 이동기능을 억제하고 수지상세포의 항원제시 기능 조절을 통하여 면역 조절에 기여할 가능성이 있음을 나타낸다.;Mesenchymal stem cells (MSCs) are one of the bone marrow (BM) stromal cells that are known to support hematopoiesis and organ recognition. Recently it has also been recently suggested that MSCs display immunosuppressive capacities through inhibiting the differentiation of monocyte-derived dendritic cells (DCs). In this study, MSCs were identified by flow cytometry of the immunologic markers and by performing colony forming unit-fibroblast assay. BM cells were isolated and cultured for the generation of myeloid DCs in the presence of GM-CSF and/or lipopolysaccharide with or without MSCs. Co-culture of MSCs inhibited the initial differentiation of DCs as well as their maturation of DCs. We addressed the effect of MSCs on the chemotactic activity of DCs in a mouse model, which one of the typical characteristics upon DCs maturation. Migration of DCs was observed via the newly developed time-lapse video microscopic technique. DCs travel to the lymph nodes (LNs) to present antigen to T cells, and CCL21 is the chemokine that plays an important role in DCs migration into the T cell area of LNs. The matured DCs actively migrated directionally in response to CCL21, whereas the MSCs co-cultured DCs did not. These in vitro observations raise the possibility that MSCs suppress the migratory function of DCs and so they may serve immuneregulatory activities through the modulation of the antigen-presenting function of DCs.
