Abstract
Chapter 1 본 연구에서는 북미대륙에 분포하는 귀뚜라미 Gryllus vernalis로부터 다형성이 있음이 확인된 5개의 microsatellite 마커를 개발했으며, 근연종인 G. fultoni에서도 사용할 수 있음을 확인했다. Micsosatellite 마커의 개발에는 pyrosequencing 방법을 사용하였다. 이 두 종은 서식지가 일부 겹치며, 형태적 차이는 거의 없고 울음소리로 구분할 수 있다고 알려져 있다. 이형접합성의 관찰값의 평균은 G. fultoni에서 0.578, G. vernalis에서 0.489이며 하디-와인버그의 평형은 개발한 micsosatellite 마커 중 M, O2에서 비교적 많은 집단에서 만족되었다. 그러나, 테스트에 사용한 개체가 여러 집단에서 채집되었고, 각 집단의 개체수도 소수였기 때문에 개체수를 늘려 추가적인 마커의 특성 분석이 요구된다. 개발된 microsatellite 마커는 두 종간의 집단 유전학적 구조의 연구에 사용될 수 있을 것이다. Chapter 2 북미대륙에 서식하는 귀뚜라미 두 종 Gryllus fultoni와 Gryllus vernalis (Orthoptera: Gryllidae)는 북미대륙 동쪽에 넓게 분포하며 서식지를 상당부분 공유하고 있다. 이 중, 동소 지역(Sympatry)에 서식하는 G. fultoni는 수컷의 울음소리와 암컷의 짝짓기 식별 행동이 이소 지역(Allopatry)과 유의한 차이를 보이며 생식적 형질 대치(Reproductive character displacement, RCD)의 패턴을 보인다고 알려져 있다. 이런 G. fultoni의 행동 패턴의 흔적을 집단 유전학적 변이 분포에서도 찾아볼 수 있을지를 알아보고 두 종의 집단 유전학적 구조를 알아보는데 연구의 초점을 맞추었다. 분석에는 microsatellite 5개 좌위와 미토콘드리아 COI 유전자가 사용되었다. G. vernalis는 microsatellite 마커를 사용한 분석과 미토콘드리아 COI 유전자 분석 모두에서 동소 지역 집단과 이소 지역 집단 간에 약한 유전적 구조가 존재하는 것으로 관찰 되었다. G. fultoni는 microsatellite 마커 분석에서는 집단 간의 유전적 분화가 관찰되지 않았으나 미토콘드리아 COI 유전자 분석에서는 동소지역과 이소지역 집단들 간에 큰 차이를 보였다. microsatellite 마커 분석에서 G. fultoni의 RCD 패턴의 흔적을 찾을 수 없었던 것은 본 연구에서 사용한 마커들이 형질 대치와 같은 자연 선택의 영향을 받는 유전자 좌위와는 독립적이라고 추측할 수 있다. 미토콘드리아 유전자 분석 결과와의 차이는 핵 유전자와 소기관 유전체의 유전적 특성 차이에서 기인된 것으로 생각한다. G. vernalis가 G. fultoni와는 달리 microsatellite 마커 분석에서도 그룹 간의 분화가 관찰되는 것은 빙하기 이후 남에서 북으로 분포 지역의 확장을 거치며 격리가 일어난 해당 종의 진화역사를 반영한 것으로 추정된다. 결론적으로 본 연구의 유전적 좌위들은 북미대륙 귀뚜라미 두 종 간 형질대치에 관여하지는 않지만 빙하기를 전후로 일어난 종의 진화역사에 대한 정보를 제공한다고 볼 수 있다.;Chapter 1 This study developed five microsatellite markers that were confirmed to have polymorphism from the field cricket G. vernalis in North America and were cross-amplified in G. fultoni, as well. Used in the development of microsatellite markers was pyrosequencing. The two species overlap in some of their habitats, and can be apart with the male calling songs. The mean values of observed heterozygosity of G. fultoni and G. vernalis were 0.578 and 0.489, respectively. The HWE equilibrium was loci M, and O2 was observed in relatively many groups. Since the individuals used in the study were collected from various regions and the number of individuals used for each region was small, there needs to analyze the characteristics of additional markers by increasing the number of individuals. The developed microsatellite markers are significant in that they will contribute to the molecular phylogenetic relations between the species and the population genetic structure of each of the species. Chapter 2 Two cricket species of North America, Gryllus fultoni and Gryllus vernalis occur in the eastern North America and share a good amount of habitats. G. fultoni in area of sympatry is significantly different from those of allopatry in terms of the male's calling song and the calling song discrimination of the female, showing a pattern of reproductive character displacement(RCD) pattern. This study focused on whether the evidence of such behavioral patterns of G. fultoni could be found in the genetic markers in addition to the group genetic structures of the two species. Used in the analysis were five microsatellite loci and mitochondria COI genes. As a result, G. vernalis showed a weak genetic structure between the sympatric and allopatric populations both in the microsatellite and mitochondrial COI gene analyse. As for G. fultoni, no genetic differentiation was observed the microsatellite marker analysis results, but a high degree of population differentiation was found in the mitochondria COI gene analysis. No genetic evidence of RCD pattern in G. fultoni were found according to the microsatellite marker analysis results probably because neutral markers used in present study are not much affected by selection such as character displacement. The differences according to the mitochondria gene analysis also resulted from the marker characteristics. Differentiations between the groups of G. vernalis were observed unlike G. fultoni according to the microsatellite marker analysis results probably because it might have gone through range expansion from south to north and isolation after the ice age. The study found no clear genetic evidence to prove character displacement in the two cricket species of North America and showed differences in analysis results according to the marker characters, thus raising a need for analysis from a different research.
