Abstract
천연 상태 식품의 품질을 유지할 수 있는 새로운 非熱 살균 기술로써 기대되고 있는 Intense Pulsed Light(IPL) 기술을 이용하여 연어회, 광어회, 새우 등의 수산 식품을 유통하는 과정에서 문제시 되고 있는 각종 위해 세균을 효과적으로 저감화할 수 있는 방법을 연구하였다. IPL 기술의 가장 근본적인 원리는 전 파장의 강한 빛을 아주 짧은 시간 안에 식품에 가하여 식품의 표면을 살균시키거나 표면 미생물 수를 감소시키는데 있다. 또한 IPL 기술은 제품의 유통기한을 연장하고 품질을 높이기 위한 목적으로 사용되며 식품 표면의 살균 뿐만 아니라 포장재나 투명한 약품의 살균에도 사용될 수 있는 유용한 기술이다. 본 연구에서는 해산물에 존재하는 위해 세균을 동정하여 위해 세균의 불활성 현상과 IPL 기술의 상업적 살균 효용성에 대하여 연구하였다. 실험결과, 해산물에서 Escherichia coli O157:H7, Listeria monocytogenes, Pseudomonas aeruginosa, Bacillus cereus 를 검출할 수 있었고, 이 균들을 대상 균주로 IPL 처리를 한 결과 전기력의 세기가 증가할수록, 처리 시간이 상승할수록 미생물의 사멸율이 증가하였다. 광펄스 1pulse 당 에너지 밀도는 전기력 세기 10, 15, 20, 25 kV triggering voltages에 대하여 각각 0.56, 1.56, 3.48, 8.75 mJ/cm2 을 나타내었다. E. coli O157:H7 은 1.56 mJ/cm2, 450 μs 에서 3 log, 8.75 mJ/cm2 의 에너지 밀도에서 750 μs 처리시 6 log의 사멸율을 나타냈었고, L. monocytogenes 의 경우 1.56 mJ/cm2, 450 μs 처리시 1 log, 8.75 mJ/cm2 에너지 밀도에서 900 μs 처리시 6 log의 사멸율을 나타내었다. 또한 P. aeruginosa 역시 8.75 mJ/cm2, 800 μs 처리시 5 log 정도의 사멸율을 나타내었으며, B. cereus는 8.75 mJ/cm2, 750 μs 처리시 6 log 정도의 사멸율을 나타내었다. 연어회, 광어회 그리고 새우에 E. coli O157:H7, L. monocytogenes, P. aeruginosa 균주를 접종하여 1pulse 당 8.75 mJ/cm2 에너지 밀도에서 IPL처리를 하여 살균효과를 살펴보았다. 실험 결과, 균주는 각 sample에서 6000 μs 처리시 1~2 log 정도의 사멸율을 나타내어 IPL 처리에 대한 해산물의 살균 효용성을 확인 할 수 있었다. 광펄스 처리 전과 후 식중독균의 내부 변화를 관찰하기 위해pulse 당 8.75 mJ/cm2 에너지 밀도에서 처리 후에 세포의 형태를 투과 전자현미경(TEM)으로 관찰하였다. 실험 결과, 광펄스를 처리 하지 않은 세포는 세포내 물질이 고르게 존재하였으나, 광펄스 처리를 한 세포에서는 세포질(cytoplasm)과 세포벽 (outer membrane) 사이의 간격이 생기고, 공간이 눈에 띄게 커져 있었다. 이러한 손상으로 인해 세포가 터지고 세포내 물질이 밖으로 유출되는 모습이 관찰되었다. 이는 세포벽이 상대적으로 더 얇은 그람 음성균의 경우 더욱 뚜렷히 나타났다. 이를 통해 광펄스의 사멸 기작으로 가장 잘 알려진 핵산 파괴 이외에 세포질과 세포벽 파괴로 인한 균의 불활성화의 가능성을 살펴 볼 수 있었다. 이와 같은 실험 결과를 바탕으로 해산물 뿐만 아니라 육류, 야채 등의 신선 식품과 영ㆍ유아용 식품 등 비가열 식품들에게서 많은 논란이 되고 있는 위해 세균에 대한 적절한 살균방법으로써 intense pulsed light 기술을 기대해 본다.;Escherichia coli O157:H7, Listeria monocytogenes, Bacillus cereus and Pseudomonas aeruginosa are representative microorganisms which can be detected in sea foods and may cause serious disease. The purposes of this study were to determine the inactivation effects of the intense pulsed light on E. coli O157:H7, L. monocytogenes, B. cereus and P. aeruginosa which are representative pathogenic microorganisms on the surface of sea foods such as flatfish, salmon, shrimp to evaluate the possibility of the commercial feasibility of this sterilization method. Overall results showed that the inactivation of microorganisms increased with increasing energy density and treatment time. The cell reduction of E. coli O157:H7 was obtained 1 log for 50 μs at 8.75 mJ/cm2 and 4 and 6 log for 300 μs and 750 μs of treatment time, respectively. The inactivation of L. monocytogenes also increased with increasing light intensity and retention time. In case of L. monocytogenes, the cell reduction was obtained 4 log for 300 μs at 8.75 mJ/cm2 and 6 log for 900 μs of treatment time. For B. cereus, the cell reduction was obtained 2 log at 8.75 mJ/cm2 and 150 μs of treatment time, 4 and 6 log for 300 μs and 750 μs of treatment time, respectively. In case of P. aeruginosa, the cell reduction was obtained 3 log at 8.6 mJ/cm2and 50 μs of treatment time, 5 and 6 log for 450 μs and 750 μs of treatment time, respectively. To study feasibility of IPL on sea foods inoculated microorganisms were inoculated on flatfish, shrimp fillet and treated by IPL. The culture fluid of E. coli O157:H7, L. monocytogenes and P. aeruginosa was inoculated by spreading on the surface of fish fillets and treated with the IPL under the following conditions: 8.75 mJ/ cm2 per pulse of energy density; 5 Hz of pulse frequency; 6000 μs of treatment time. The inactivation of microorganisms increased with increasing treatment time. The cell reduction of E. coli O157:H7 was obtained up to 1 log on salmon fillet, 1.5 log on flatfish fillet and 2 log on shrimp fillet. In case of sea foods inoculated with L. monocytogenes, the results showed that the cell reduction was obtained 1, 1.2 and 1.5 log on flatfish, salmon and shrimp fillet, respectively. For sea foods inoculated with P. aeruginosa, the results showed that the cell reduction of P. aeruginosa was obtained 1.3, 1.5 and 1.8 log on flatfish fillet, salmon fillet and shrimp fillet, respectively. To evaluate the cell damage by IPL treatment processes, microorganisms treated by IPL were observed by a transmission electron microscopy. The microscopic observation clearly indicated that there was cell wall damage, cytoplasmic membrane shrinkage and cellular content leakage.
