조절방출성 신제제를 위한 소프트하이드로겔에 대한 연구

Abstract

생체 적합성, 약물 방출 조절 기능이 우수하고 물성이 특이하여 조절 방출성 신제제로 광범위하게 활용될 수 있는 소프트하이드로켈의 개발을 위한 연구를 수행하였다. Poly(alkylene oxides)가 triisocyanate로 crosslinked되고 diisocyanate로 extended된 hydrogel를 제조하였다. 친수성/소수성 모델 약물을 선정하여 crosslinker와 extender 비율 조절에 따른 hydrogel의 약물 방출 조절능을 조사하였으며, 팽윤 실험을 실시하여 약물 방출 기전을 검색하고자 하였다. 또한, 제조된 소프트하이드로겔의 본질적 우수한 물성을 유지하면서 조절 방출에 필요한 특수기능성을 부가시키고자 기능성 고분자류인 polyacrylate류와 IPNs 공중합체를 합성하여 수팽윤성, pH 의존성 등의 물성을 조사하였으며, 비이온성/양이온성/음이온성 모델약물을 선택하여 hydrogel의 pH에 따른 가변적인 팽윤양상과 이온성 상호작용에 근거하여 약물 방출 조절 기전을 규명하고자 하였다.; Hydrogels are widely studied materials for the preparation of controlled release drug dosage forms. Their soft, tissue-like consistency and their high biocompatibility in a number of applications make them promising candidates for this purpose. Firstly, poly(alkylene oxides) was crosslinked by diisocyanates and triisocyanates to form physically strong, rubber-like elastic, high biocompatible polyurethanes. The isocyanate-hydroxyl stoichiometry was kept at 1:1, but the crosslink density was varied. The variations examined were the ratio of diisocyanate and triisocyanate. Various preparation parameter influenced the delivery rate by means of a modified effective mesh size of the network. The release profiles could be modified to a great extent by adjusting the polymer network structure. Secondly, an interpenetrating polymer networks consists of two chemically independent crosslinked polymers in which the proportion and properties of both polymers can he independently varied. The first prepared network is a cr-PEG and the other consists of or-PAA. These IPNs show excellent pH dependence of swelling behaviors and controlled release of drugs dependent on ionic intermolecular interaction.
