Abstract
Stress is one of crucial factors which induce common clinical problems such as anger, anxiety and depression. Especially depression is known as being directly caused by stress. However, the precise mechanisms of neuronal injury by stress are still unclear. This study investigated whether the neuronal damage by two kinds of stresses such as immobilization and corticosterone injection is induced by anxiety and modulation of the expression of TH and pERK. In addition, the anxiolytic effect of TMCA derivatives was examined in stress model. Firstly, the present study identified the neuronal damage by 10 minute consecutive electric shock stress by using elevated plus maze and measuring the expression of catecholamine enzyme, tyrosine hydroxylase and mitogen-activated protein, pERK. The rats exposed to stress showed anxiety since spent lesser time in the open arms than the control groups. Stress led to the decrease of the expression of TH and increase of the expression of pERK in amygdala but not in cortex. To identify the effects of corticosterone, which is increased by stress, rats were injected corticosterone for 3-week consecutive days. This treatment also showed anxiety and reduced the expression of TH in amygdala. These results indicated that the two kinds of stress models, electric shock and corticosterone injection, accorded with induction of anxiety and modulation of TH. To confirm the biomolecular effect of corticosterone in stress, the levels of iNOS, GFAP and pERK were ascertained in cortical neuronal cells and BV2 microglial cells. The expression of iNOS and pERK were elevated and the expression of GFAP was decreased both in corticosterone-treated neuronal cells and microglial cells. Secondly, the anxiolytic effects of TMCA derivatives were estimated in stress model. Compound TMCA derivatives were synthesized and administered to intraperitoreally at 30 min prior to stress. Pre-treatment with TMCA derivatives before stress, reduced anxiety while with stress without treatment alone increased anxiety. In addition, the TH activities were decreased by the treatment with TMCA derivatives. To identify the anxiolytic mechanisms of TMCA derivatives, dopamine and serotonin receptors binding assay was performed since it was known that TH activity was involved in stress and serotonin system plays importantly in depression and anxiety. TH is a rate limiting enzyme in the synthesis of catecholamine that is activated by stress stimulus. TMCA derivatives show high binding affinity on the serotonin receptors, especially on the 5-HT1A receptor. Thus, these results might suggest that TMCA derivatives have anxiolytic effects by modulation of TH and pERK via 5-HT1A receptor. Collectively, this study suggests that repeated stress-induced anxiety may modulate the expression of TH and pERK in amygdala. TMCA derivatives may activate the 5-HT1A receptor and could be applied to useful anxiolytic agents for the treatment of stress.;스트레스는 화(anger), 불안(anxiety) 혹은 우울(depression) 과 같은 증상을 야기시키는 중요한 요소중의 하나이다. 특히 우울증은 만성스트레스로부터 직접적으로 발생된다고 알려져 있다. 그러나 스트레스가 뇌에 어떤 영향을 주며, 그 구체적인 작용 기전이 무엇인지는 여전히 불명확하다. 본 논문에서는 스트레스 모델을 사용하여 편도체와 대뇌피질에서의 스트레스 반응에 의해 발현이 달라지는 신호관련 단백질 발현을 관찰하였으며, 콜티코스테론이 스트레스 시 증가되는 것을 확인한 후 콜티코스테론을 직접 처치 시 변화되는 불안행동측정과 신호관련 단백질 발현의 변화를 관찰하였다. 또한 TMCA의 유도체를 만들어 스트레스 모델에서의 동물불안행동 측정과 단백질 발현변화를 통하여 항 스트레스 효과가 있음을 확인하였다. 스트레스 모델 확립을 위하여 부동 및 전기자극스트레스를 준 후에 동물불안행동 측정과 단백질의 발현을 측정한 결과 전기자극을 받은 동물에서 불안 증세가 나타났으며 스트레스군이 대조군에 비해 편도체에서 Tyrosine hydroxylase(TH)와 같은 catecholamine 생성효소의 발현을 감소시키며, pERK의 발현을 증가시키고 혈중 콜티코스테론이 증가함을 확인할 수 있었다. 스트레스 시 증가되는 콜티코스테론의 뇌 내 작용을 규명하고자 콜티코스테론을 3주간 매일 주사 후, 동물불안행동 측정 및 단백질의 발현을 확인해 보았다. 그 결과 콜티코스테론을 처리한 군은 대조군에 비해 교가십자로(elevated plus maze)에서 open arms에 머무는 시간이 감소하였고, 대뇌피질에서의 발현은 크게 변화가 없었지만 편도체에서의 TH 및 pERK의 발현양은 감소하였다. 이는 콜티코스테론이 불안을 유발함을 알 수 있었다. 또한 콜티코스테론이 초대 배양 대뇌 전후 세포(primary cortical neuronal cell)및 아교세포(microglia)인 BV2 cell에서 어떤 영향을 주는지 알아보기 위해, 콜티코스테론 처리 후 세포 내 신호 단백질 발현을 관찰하였다. 콜티코스테론을 처리한 세포에서 NO의 합성에 관여하는 iNOS, 교세포 활성화에 관여하는 GFAP 그리고 세포의 증식과 분화에 관여하는 ERK의 발현을 규명한 결과 iNOS의 발현은 증가 하였으며 GFAP와 ERK의 발현은 감소하여 콜티코스테론이 신체구금 스트레스 상황과 유사한 결과를 유발하는 것을 확인할 수 있었다. 다음으로, 스트레스 억제에 대한 TMCA의 유도체의 효과를 규명하기 위하여, 신체구금과 전기자극을 함께 가하여 스트레스를 유발시키며 TMCA 유도체를 병용 투여한 결과 동물불안행동 측정에서 스트레스를 완화시키는 효과를 보였다. TMCA 유도체의 항 스트레스에 대한 약리학적 작용기전을 규명하기 위하여 도파민 및 세로토닌 수용체에 대한 수용체 결합 실험을 수행한 결과 흥미롭게도 대부분의 TMCA 유도체는 세로토닌 수용체 중 5-HT1A 수용체에 높은 결합력을 나타내었다. TMCA유도체가 세로토닌 수용체 중 5-HT1A에 결합하여 항스트레스 효과를 보이는지를 규명하고자 5-HT1A 수용체의 선택적인 agonist 8-OH-DPAT 을 처치한 뒤 동물불안행동 측정과 단백질의 발현을 확인 본 결과 불안이 감소 하였으며 TH의 발현 또한 증가함을 알 수 있었다. 본 연구의 결과는 전기자극 및 반복된 콜티코스테론 주입에 의한 불안 유발모델 구축에 의미가 있으며 콜티코스테론이 불안유발 스트레스 연구에 유용함을 입증하였다. 또한 합성된 TMCA 유도체가 뛰어난 항 스트레스 효과를 나타내며 이런 작용기전은 세로토닌 수용체에 결합함으로써 항불안 작용을 나타냄을 규명하여 TMCA 유도체가 항스트레스 치료에 응용될 가능성을 규명하였다.
