Solvent deposition method를 利用한 indomethacin의 溶出速度의 增加에 관한 硏究

Abstract

To evaluate the pharmaceutical aspects of solvent deposition method where drug is solvent deposited on the surface of excipients, a study has been made on dissolution characteristics of indomethacin solvent deposited on lactose and potato starch. In a solvent deposition system, the drug-to-excipient ratio and kind of excipierit effect much on dissolution rates of indomethacin. The experimental results are as follows: 1) Lactose was shown to be superior to potato starch as excipients in indomethacin solvent deposited. 2) Total amount of indomethacin dissolved from solvent deposition systems at 30 minutes were enhanced about 5 to 23 times compared with that of pure indomethacin. 3) Increased dissolution amount of indomethacin from the solvent deposition systems were observed to be alike in the systems where the drug-to-excipient weight ratios were 1:5, 1:7 and 1:10.;難溶性藥物인 Indomethacin을 Solvent Deposition Method를 이용하여 賦形劑인 Lactose와 Potato Starch 表面에 沈着시킨 Solvent Deposition System의 溶出特性을 藥劑學的인 面에서 관찰하였다. Solvent Deposition의 영향을 세가지 다른 溶出溶媒에서 Lactose와 Potato Starch를 使用하고 醫藥品과 賦形劑의 比를 변화시켜 만든 Solvent Deposition System의 溶出速度를 비교 검토하여 다음과 같은 結果를 얻었다. 1) Indomethacin의 Solvent Deposition System에서는 賦形劑로서 Potato Starch보다 Lactose가 더 우수하다. 2) 溶出時間 30分을 기준으로 했을 때 溶出量은 Control에 比해 5∼23倍로 증가하였다. 3) 醫藥品과 賦形劑의 比는 1:5, 1:7, 1:10인 경우 거의 같은 溶出量을 보였으며 市販 Indomethacin Capsule 劑와 同一한 1호 Hard Capsule에 충전할 때는 1:5가 가장 적합하였다.
