Abstract
Promethazine은 Phenothiazine 유도체의 일종이다. Promethazine pamoate는 Promethazine-HCl의 약물효과를 지속시키고 독성을 저하시키기 위해 Laboratories Sobio, S. A. France에서 개발했다. 著者는 Promethazine pamoate의 합성 및 분석에 관해 연구하였다. Promethazine pamoate는 Pamoate acid 1 mole과 Promethazine-HCl 2 mole로 구성되며, Pot. Pamoate 용액을 Promethazine-HCl에 서서히 가하여 침전을 형성시키며 이때의 pH는 5.0이다. Promethazine PdCl₂ 발색시약과의 정색반응은 최대 흡수파장 475nm에서 측정하며 Promethazine pamoate 제제의 정량분석에 이용된다. Promethazine의 형광분석법은 60℃에서 10분간 가온하여 산화시킬때 생성되는 형광을 측정하는 것으로 Excitation Have length 300nm, Emission Wavelength 400nm에서 측정한다.;Promethazine belongs to phenothiazines. Prone thazine pamoate was developed from Laboratories Sobio, S.A. France in order to maintain long acting antitussive activity end decrease the toxicity of promethazine. We researched the effective method of synthesis and quantitative analysis of promethazine pamoate. Promethazine pamoate was obtained by treatment of a mol pamoic acid with 2 moles promethazines base in a suitable solvent, where pottasium pamoate solution was added slowly to promethazine hydrochloride solution at PH 5.0 The color reaction between promethazine and palladiem chloride was applied to the determination of promethazine pamoate, and the color solution exibited absorption miximum at 475nm. This colorimetric determination method could be used as the quantative analysis of promethazine pamoate preparation. The fluorometric determination of promethazine was based upon the fluorescene of vromethazine which occured after heating for 10 minutes at 60℃, and the fluorescent spectra had excitation maximum at 300nm and emission maximum at 400 nm.
