Abstract
CETP (Cholesterol ester transfer protein)는 혈액내의 CE (cholesterol ester)의 운반과정에 관여하는 효소로서 cholesterol을 체외로 제거하는 유일한 기작인 reverse cholesterol transport pathway의 cholesterol의 흐름을 조절해주는 율속효소이다. CETP는 과량의 cholesterol을 핏속에 오랜 시간 머무르게 하며, 그 결과 cholesterol을 필요이상으로 섭취하는 현대인에게 동맥경화증을 비롯한 각종 심혈관계 질환의 원인을 제공한다고 알려져 있다. 즉, CETP발현이 없는 쥐, 돼지와 같은 동물실험 및 최근 일본에서 발견된 한 CETP겹핍가계에서 동맥경화증에 대한 강한 저항성이 나타난다고 보고되었으나 CETP를 발현시킨 쥐에서는 관상동맥 내면에 침착되는 거품세포의 형성이 현저히 증가됨으로서 동맥경화증을 악화시킬 수 있는 요인으로 작용하고 있다. 따라서 선진각국을 중심으로 CETP저해제 개발이 활발히 진행되고 있으나, 현재까지 in vivo활성이 보이는 CETP저해제를 찾지 못한 상황이다. 본 연구에서는 생약중 CETP저해활성을 가진 물질을 찾고자 천연물을 대상으로 검색하던 중 창이자 (도꼬마리 Xanthii Fructus)에서 저해 활성성분이 있음을 확인하였다. 도꼬마리는 국화과 (Compositae)식물로 우리나라에서는 Xanthium strumarium L.이 중국에서는 Xanthium sibiricum이 사용된다. 성숙한 과실은 창이자(蒼耳子, Xanthii Fructus)라 하여 비염, 축농증 등에 약용되었다. 창이자는 혈당강하작용이 있고, 전제는 항균작용이 있으며 유독하고, 해열, 발한, 진통, 산풍, 거습, 궤양성 피부병, 신경통 및 악성종양에 효과가 있다. 창이자에서 CETP저해활성성분을 검토한 결과, compound I (β-sitosterol g1ucoside), Compound Ⅱ (C_(18)H_(27)NO_7)S), compound Ⅲ (3-(2-Hydroxy-4-me- thoxyphenyl)-2-propenoic acid)를 분리하였다. compound Ⅱ, Ⅲ의 CETP에 대한 IC_(50)는 각각 50㎍/㎖, 100 ㎍/㎖였다. 보다 우수한 효과를 보인 compound Ⅱ에 대해 그의 acetate를 합성하여 CETP에 대한 저해활성을 측정한 결과, IC_(50)가 40 ㎍/㎖로 CETP저해활성이 강화되었다. 창이자에 대한 CETP활성은 현재까지 검토된 바 없었다. compound I은 본 식물에서 이미 보고된 적이 있는 화합물이며, CETP저해활성은 없었다. Compound Ⅲ는 Artemisia anomala를 비롯, A dracunculus등 국화과 (Compositae)식물에서 보고된 바 있으나, 창이자에서는 처음 분리되었고, CETP저해활성에 관해서도 보고된 바 없었다. 또한 compound Ⅱ는 현재까지 밝혀진 분자식으로 볼 때, 천연물에서 흔히 볼 수 없는 신 물질로 추정되며, CETP저해활성에 관해서도 보고된 바 없었다. 이상의 사실로 미루어 볼 때, CETP 저해활성을 보인 compound Ⅱ, compound Ⅱ의 acetate, compound Ⅲ는 고콜레스테롤혈증과 동맥경화증의 치료에 효과적으로 이용될 가능성이 있는 것으로 사료된다.;Atherosclerosis is one of the leading causes of death in western industrialized countries. Diagnosis, prevention, and treatment of atherosclerosis are not sufficient so far. The clinically used antiatherosclerotic agents, HMG CoA reductase inhibitors, lovastatin and simvastatin, bile acid-binding resins, cholestyramine resin, fibric acid derivatives, clofibrate probucol and nicotinic acid, have many adverse effects, diarrhea, pruritus, bloating, constipation, etc. Therefore more safe and effective agents are eagerly needed. CETP, cholesterol ester transfer protein, is a lipid transfer protein, which mediates the transfer of cholesterol ester and triglyceride between high-density lipoprotein (HDL) and other low-density lipoprotein (VLDL, LDL). It is reported that plasma HDL level decreased in human CETP gene transgenic mouse and increased in genetic CETP deficiency human. Increase of anti-atherogenic HDL decreased the development of atherosclerosis. These reports suggest that elevation in CETP activity strongly relates to the development of atherosclerotic cardiovascular diseases. In the course of screening CETP inhibitors from Korean traditional medicines, the methanol extract of Xanthii Fructus was showed 10% inhibition effect in CETP assay system at 100 ㎍/㎖, significantly. The three compounds are isolated from this plant by using silicagel column chromatography, RP-C_(18) column chromatography and HPLC. Of the isolated three compounds, compound Ⅱ, and compound Ⅲ showed CETP inhibitory activity, IC_(50)=50, and 100 ㎍/㎖, respectively. Compound Ⅱ-acetate showed stronger activity than compound Ⅱ, IC_(50)=40 ㎍/㎖. The structures of the isolated compounds were elucidated as compound I, β-sitosterol glucoside and compound Ⅲ, 3-(2-Hydroxy-4-methoxyphenyl)-2-propenoic acid by using UV, IR, Mass, NMR spectroscopy. The molecular formula of compound Ⅱ was prospected C_(18)H_(27)NO_(7)S and its structure is under elucidation.
