- 수용성 약물인 Propranolol-HCI(PPH)을 Model Drug으로 하여 Ethylcellulose(EC), Methylcellulose(MC), Hydroxypropylcellulose(HPC), Hydroxypropy1-methylcellulose(HPMC)등의 Polymer를 사용하여 Solid Dispersion System(SDS)을 제조하여 용출특성을 검토하고 수용성 약물의 지속성 제제의 개발을 목적으로 본 시험을 시행하였다.
PPH와 Polymer를 absolute alcohol에 용해 또는 현탁시킨 Solvent Method로 SDS를 제조하여 적외 흡광 광도법 및 분말 X-ray회절빔으로 물리화학적 성질을 관찰하고 용출 시험을 시행하였다.
PPH-EC-SDS와 PPH-MC-SDS는 분말 X-ray 회절시험 결과 PPH의 결정성 감소를 확인 할수 있었으며 IR-Spectra 상에서도 PPH에 의한 Peak의 Intensity가 감소하거나 혹은 둔화되는 양상을 보인다.
용출 실험 결과 PPH-MC-SDS, PPH-EC-SDS이 지속성을 나타내었다. PPH-MC-SDS는 PPH-EC-SDS에 배해 초기 용출 속도가 완만하였으며 T 50%도 PPH-EC-SDS의 40분데 비해 5시간으로 현저히 지연되었다. 그리고 PPH : MC의 비가 1:5,1:7일때 Square Root Time에 대하여 직선성을 나타내었다.
또, PPH-MC-SDS와 PPH-EC-SDS의 용출 양상은 용출 용매의 pH에 영향을 받지 않았고, 입자경이 적을수록 용출속도가 빨랐다.;The object of this study was to develope of sustained release dosage form of water soluble drug by Solid Dispersion System ( SDS ) which was prepared by Solvent Method with Propranolol-HCl ( PPH ) as a model drug and also with polymer such as Methylcellulose ( MC ), Ethylcellulose ( EC ), Hydroxypropylcellulose ( HPC ) and Hydroxypropylmethylcellulose ( HPMC ) as Carriers.
The IR-spectra and Powder X-ray Diffraction tests were carried out for physicochemical property assessment
It was found that MC-SDS and EC-SDS exhibited sustained release pattern and showed independent release Pattern from pH in Dissolution Medium. The release rate of MC-SDS was slower than that of EC-SDS at early phase. PPH-MC-SDS ( 1:5 ) and PPN-MC-SDS ( 1:7 ) showed the linearity as the function of square root of time.
