현재 우리나라에서 허용되어 있는 식품 첨가물중 azo색소는 amaranth, tartrazine, sunset yellow 및 allura red로서 모두 분자중에 술폰산기(-SO_(3)H) 및 수산기(-OH)를 가지고 있다. 이러한 수용성의 산성 azo색소가 간 microsome내에서 많은 물질의 활성화와 해독작용을 나타내는 주요 대사과정중 전자전달계 효소인 mixed function oxidase system에 미치는 영향을 알아보고자 각 azo색소들을 80mg/kg으로 흰 쥐의 복강 내에 1일 1회 4일간 투여하고 간 microsome을 분리하여 cytochrome P-450, cytochrome b_(5), NADPH cyt. c-reductase 및 azo reductase의 활성을 측정하였다. 또한 효소 상호간의 의존성을 좀 더 구체적으로 구명하고자 간의 물질대사 효소를 유도시키는 것으로 알려진 phenobarbital 및 3-methylcholanthrene과 물질대사 효소의 합성을 저해하는 것으로 알려진 cobaltous chloride 및 carbon tetrachloride를 각각 투여하고 cytochrome P-450 및 NADPH cyt. creductase 활성에 따른 azo reductase의 활성을 측정하여 다음과 같은 결과를 얻었다. 1. Sulphonate azo 색소는 hepatic microsomal Cyt. P-450 및 Cyt. b_(5)의 함량을 감소시켰다. 2. Sulphonate azo 색소는 hepatic microsomal azo reductase의 활성을 감소시켰으며, 감소된 양상은 Cyt. P-450함량의 감소양상과 동일하였다. 3. Sulphonate azo색소는 NADPH cyt. c-reductase 활성에 영향을 미치지 않았다. 4. Azo reductase활성은 3-methylcholanthrene 투여에 의해서도 증가되었다.;The four azo dyes such as Amaranth (FD & C Red No.2), Tartrazine (FD & C Yellow No.4), sunset Yellow (FD & C Yellow No.5) and Allura red (FD & C Red No.40) are currently employed as a food additives in Korea. In this study, the effects of these azo dyes on the hepatic microsomal mixed function oxidase systems in Rats. (i.e., Cyt. P-450, Cyt. b_(5), NADPH cyt. c-reductase and azo reductase) were investigated. Furthermore, to determine the relationship among the electron transport systems, each level of azo reductase, Cyt. p-450 and NADPH cyt. c-reductase were measured upon the administration of phenobarbital (known as an inducer of Cyt. p-450), 3-methylcholanthrene (Known as an inducer of Cyt. p-448), CoCℓ_(2) (inhibitor on Cyt. p-450) or (CCℓ_(4) (inhibitor on Cyt. p-450). The results of these studies are as follows; 1. The level of Cyt. p-450 and Cyt. b_(5) were decreased upon the administration of these azo dyes. 2. When the level of Cyt. p-450 was decreased, the azo reductase activity was also decreased. 3. These azo dyes did not show any significant effect on the level of NADPH cyt. c-reductase. 4. The administration of 3-methylcholanthrene resulted in the elevation of azo reductase activity. The 3-methylcholanthrene may be responsible for the induction of CO-insensitive electron transport system.
